These systems force nucleic acids to bind to silica under high salt and ethanol concentrations—followed by multiple washing and elution steps. This method is indirect, harsh, and inefficient: not only are valuable nucleic acid fragments lost during binding or washing, but the method is also time-consuming and labor-intensive, requiring numerous high-speed centrifugation steps and consumables.
