BioEcho's EchoLUTION FFPE RNA Kit Outperforms Competing Methods in Independent Head-to-Head Evaluation

An independent performance evaluation conducted by TAmiRNA, a specialist in small RNA biomarker research, has ranked BioEcho's EchoLUTION FFPE RNA Kit as the top-performing solution for RNA isolation from formalin-fixed paraffin-embedded (FFPE) tissue — ahead of both the silica-membrane extraction and magnetic bead-based method.

FFPE samples are among the most challenging starting materials in molecular diagnostics. Formalin fixation causes chemical modifications and cross-linking that degrade RNA and make recovery difficult. Despite this, high-quality RNA extraction from archival FFPE tissue is essential for oncology research, biomarker discovery, and clinical pathology workflows.

The evaluation compared all three kits across RNA yield, RNA quality, cDNA library quality, and next-generation sequencing (NGS) output. Across these criteria, the EchoLUTION FFPE RNA Kit achieved the highest combined score of 11.5 out of 12, compared to 8.75 for the silica-membrane extraction kit and 8.0 for the magnetic bead-based extraction kit.

While RNA yields were broadly similar across all three methods, the EchoLUTION kit stood out in sequencing performance. Extracts produced with EchoLUTION showed the highest percentage of miRNA reads, the lowest proportion of rRNA background, and detected the greatest number of distinct individual miRNAs. In the miRNA profiling comparison, EchoLUTION and silica-membrane extraction method showed highly concordant miRNA expression profiles, while both detected significantly more miRNAs than the magnetic bead-based extraction method. The EchoLUTION kit also displayed the least optical degradation in electropherogram profiles — an indicator of RNA preservation during extraction.

From a practical standpoint, the EchoLUTION FFPE RNA Kit received the highest ratings for usability and speed. The kit's single-step purification approach — the hallmark of BioEcho's EchoLUTION technology — requires no binding, washing, or elution steps, allowing experienced users to move from sample to purified RNA in a fraction of the time required by conventional methods. It is also the most cost-effective option evaluated, priced below both silica-membrane extraction method and magnetic bead-based extraction kits.

Based on the evaluation results, TAmiRNA concluded that the EchoLUTION FFPE RNA Kit is recommended as the standard FFPE RNA isolation kit for future use in small RNA sequencing workflows.
