Photoaktivierte Lokalisationsmikroskopie (PALM) in 3D mit ZEISS ELYRA

Als Erweiterung des 3D-Mikrokopie Portfolios stellt ZEISS auf der Jahrestagung der Society for Neuroscience in San Diego, USA, 3D-PALM vor. Mit einem neuen Modul ermöglicht ELYRA P.1 hochauflösende photoaktivierte Lokalisations- mikroskopie (PALM) in 3D für endogen exprimierte, photoaktivierbare Fluoreszenz- proteine. Nutzer können hochaufgelöste Strukturen in 3D erfassen und probenschonend lange Zeitreihen aufnehmen.

Mit PALM werden fluoreszierende Moleküle nacheinander aktiviert, so dass nur jeweils ein Molekül in seinem eingeschalteten Zustand innerhalb einer Punktspreizfunktion (PSF) liegt. In 3D kodiert die Form der PSF die Z-Position des Moleküls. Die Positionen aus den Einzelbildern werden zu einem neuen hochaufgelösten Bild zusammengesetzt.
